There is this game which I played a long time ago. It came in one of those CDs with hundreds of game and you didn't even need to install.
The idea was basically a top-down shooter that you could play in a multiplayer fashion up to , I don't know, 4 players in the same keyboard?
You controlled human soldiers in fortresses and fought against alien spiders and their cocoons. They spawned from time to time, making it harder to win.
The graphics were cartoonish , I'm not mistaken.

Based on your info here is a list of shooters with a shared screen:

(Assuming DOS, as you did not mention the platform)

It's extremely similar to Machine Hunter, but still is not the right one.

Could it be
Threat? The game supports up to 3 players. I wouldn't call the enemies cartooney, except maybe for the 3D rendered one in the screenshots.
To get better answers, even after a year has passed, it would help if you provided more specific info about the platform (DOS/Windows) and the year you played the game. MobyGames URLs have changed, so here's a new example where all DOS top-down games between 1992 and 2002 are listed:
https://www.mobygames.com/game/from:199 ... te/page:1/. The list of course lacks a lot, but is a good place to continue.
